The implementation of quantitative expression profiling in a novel Random Encoded Array Detection (READ) format is proposed. READ, a proprietary custom bead array technology developed at BioArray Solutions, combines bead chemistry with semiconductor technology to provide a universal platform for quantitative, multiplexed DNA and protein analysis. This novel array format affords complete flexibility in the selection of constituent bead-displayed capture probes and permits "snapshot" imaging of assays using a fluorescence microscope. Here, a novel assay format is proposed that takes advantage of inherent detection sensitivity to eliminate PCR amplification while relaying on the high redundancy and adjustable array composition to accommodate the requisite four orders of magnitude in message expression levels. A compact, fieldable system implementation will be delivered. BioArray Solutions random encoded array detection format is in beta testing stages at leading medical diagnostics centers in the US, currently for multiplexed genotyping and carrier screening as well as quantitative protein profiling. The company intends to add quantitative expression profiling to this menu of capabilities and will focus on the commercial development of custom bead array for rapid quantative expression profiling for the presymptomatic diagnosis of cancer, especially leukemia and related hematologic disorders to guide selection of treatment options and monitor treatment response

The implementation of quantitative expression profiling of blood-borne pathogenes in a novel Random Encoded Array Detection (READ) format is proposed. READ, proprietary custom bead array technology developed at BioArray Solutions, combines bead chemistry with semiconductor technology to provide a universal platform for quantitative, multiplexed DNA and protein analysis. This novel array format affords complete flexibility in the selection of constituent bead-displayed capture probes and permits snapshop imaging using a proprietary Array Imaging System with a signal decoding software. BioArray Solutions proposes to develop and validate custom BeadChip array designs and assay protocols for multiplexed expression monitoring ('gene expression fingerprinting') of a set of 100 common response and disease-specific genes on an integrated assay delivery system.

Benefits:
BioArray Solutions READ system, including a proprietary Thermoprofiler and Array Imaging System to enable high sample throughput, is in beta testing stages at leading medical diagnostics centers in the US, currently for multiplexed genotyping and carrier screening as well as quantitatve protein profiling. The company is undertaking the development and commercialization of custom BeadChips for rapid sensitive and easy to use quantitative gene expression profiling in order to expand its menu of capabilities within DNA/RNA diagnostics.
